Detoxification of zearalenone produced by Fusarium graminearum by purified tannase from Citrobacter freundii

ABSTRACT
Tannase has been used extensively pharmaceuticals and chemical industries. Zearalenone (ZEN) is a myco-estrogen in animals and a phytohormone in plants mainly produced by Fusarium graminearum. It leads to serious and chronic health-damaging effect on humans and animals. The aim of this study to investigate tannase ability to inhibit zearalenone production by Fusarium graminearum.
Methods: Environmental isolates were collected from gastrointestinal tract of fish. Simple laboratory testing followed by the complementary Vitek 2 system. The screening for tannase production was done by semi-quantitative and quantitative analysis. Tannase was purified by chromatographic methods and Different concentrations of purified tannase were used to reveled the ability of tannase to remove or destruct of zearalenone produced by Fusarium graminearum.
Results: Out of 20 fish samples,4 Citrobacter freundii isolates were obtained with percentage of 20%. Among these four isolates two isolates exhibited the ability to produce tannase enzyme and Citrobacter freundii F3 gave higher tannase productivity. This enzyme was purified to homogeneity by sequential chromatographic steps involved ammonium sulfate precipitation, ion exchange on DEAE-cellulose column and gel-filtration on Sephadex G-50 with a yield of 31.2% and 15.45fold of purification. The detoxify effect of purified tannase against zearalenone produced by Fusarium graminearum showed that with increasing the concentrations of purified tannase the tannase can effectively inhibit zearalenone production in liquid medium until 500μg/ml but in the concentrations that higher than 500 μg/ml there is decreasing in the removal.
Conclusions: This is the first report in Iraq which revealed that tannase has a promising application as detoxify agent since zearalenone and related pathology has become a worldwide preoccupation and it raises serious economic and sanitary problems.
